The Protective Effect of Co-supplementation of N-acetyl Cysteine and Magnesium against Gentamicin Induced Nephrotoxicity in Albino Rat

Increased production of reactive oxygen species (ROS) may be a common pathway in atherosclerosis, diabetes, cataract, accelerated ageing, drug toxicities. The protective role of N-acetyl cysteine (NAC)and magnesium(Mg) in gentamicin induced nephrotoxicity was evaluated . 100 albino rats were divided into 5 groups. Sham control (A), Gentamicin group (B), Gentamicin + NAC group(C), Gentamicin + Mg group( D), and group E (NAC + Mg) + gentamicin. 24 hour urinary albumin excretion (UAE), serum creatinine (Scr), superoxide dismutase (SOD), catalase (CAT) reduced glutathione and malondialodehyde in plasma (MDAP) were measured.Scr ,UAE and MDAP were significantly increased in gentamicin treated groups while SOD, CAT and reduced glutathione were significantly decreased compared with those in the control group. Combination between NAC and Mg produced significant decrease of lipid peroxidation production and increase of antioxidant enzymes. Increased lipid peroxidation and decreased antioxidant enzymes may play a role in gentamicin induced renal toxicity. Combined NAC + Mg may ameliorate these changes and protect the kidney.
